CloningandSequenceAnalysisofRibosomalProteinL21Gene fromtheAiluropodamelanoleuca

RibosonalproteinL21whichisacomponentofthe60slargeribosomalsubunitplaysanimportantrole inribosome.ToexplorethestructurecharacteristicofribosomalproteinL21(rpL21)geneoftheAiluropoda melanoleucaandinvestigateitshomologieswithotheralreadyreportedsequenses’includingRattusnorvegicus, Musmusculus,Musmusculus,etc.ThecDNAofrpL21wasclonedfromtheAiluropodamelanoleucabyRT-PCR.ThesequencedatawereanalyzedbyGenscansoftware.Blast2.1wasusedtostudythehomologyoftheob-tainedrpL21sequencewiththegenesequenceofotherspecies;Openreadingframe(ORF)oftheDNAsequence wassearchedusing ORFfindersoftware;ProteinstructureoftherpL21sequencecloned wasdeducedusing PredictProteinsoftware.TheresultsindicatedthatthelengthofcDNAfragmentclonedwas504bp;containing anopenreadingframeof483bp.Deducedproteinwascomposedof160aminoacidswithanestimatedmolecular weightof18.59kDandpIof11.10.Thelengthofthegenomicsequenceis2254bp,containing5exonsand4 introns.AlignmentanalysisindicatesthatrpL21ishighlysimilaritywiththereportedspeciesbothatthelevelof DNAandprotein.Topologypredictionshowsthat6differentpatternswerefoundintherpL21proteinoftheAi-luropodamelanoleuca.TherpL21geneoftheAiluropoda melanoleuca wasstudiedinthispaper,which will enrichandimprovethemammals’rpL21genedatabase.
